E.coli HCP 產(chǎn)品性能對比分析

背景
縱觀生物藥物生產(chǎn)的歷史,從藥物終產(chǎn)物中,有效地去除和檢測HCP雜質(zhì),有著大量的研究和發(fā)現(xiàn)。這些雜質(zhì)的存在,對臨床病人的安全和藥物穩(wěn)定性有著廣泛的影響。
目前,常見的蛋白藥物表達(dá)系統(tǒng),有原核表達(dá)系統(tǒng),如Escherichia coli ,E.coli;植物表達(dá)系統(tǒng);真菌表達(dá)系統(tǒng),如畢赤酵母,釀酒酵母;昆蟲表達(dá)系統(tǒng),如Spodoptera frugiperda cells,SF9昆蟲細(xì)胞;哺乳動(dòng)物細(xì)胞表達(dá)系統(tǒng),如中國倉鼠卵巢細(xì)胞(Chinese hamster ovary cells, CHO),非洲綠猴腎細(xì)胞(African green monkey kidney cells,Vero)等。其中哺乳動(dòng)物細(xì)胞表達(dá)系統(tǒng),在臨床藥物的生產(chǎn)中,使用較為廣泛,但是價(jià)格昂貴。產(chǎn)量最高,價(jià)格低廉,用途最廣泛的是E.coli表達(dá)系統(tǒng)。
E.coli表達(dá)系統(tǒng)是目前使用領(lǐng)域最多元的表達(dá)系統(tǒng),由于其培養(yǎng)簡單,轉(zhuǎn)化效率高,成本低,產(chǎn)量高等特點(diǎn),而被廣泛應(yīng)用。諸如不需要修飾的人胰島素和生長激素,同時(shí)自然狀態(tài)下需要糖基化修飾,但可以不糖基化修飾的蛋白,如 α-,β-和 γ-干擾素,以及白介素2和腫瘤壞死因子,仍具有生物學(xué)活性[1,2]。
對HCP的檢測和鑒定方法,通常的金標(biāo)準(zhǔn)是ELISA和LC-MS法。其中,前者可以高通量定量,且價(jià)格低廉,后者可以定量定性,但是價(jià)格昂貴,且通量較低。兩者需要正交搭配使用,來對藥物中的HCP進(jìn)行定性和定量。
有較多文獻(xiàn)報(bào)道,不同藥物的相同純化工藝,或者相同藥物不同工藝的HCP殘留均有較大差別,因此,國內(nèi)外的藥物HCP定量結(jié)果不盡相同[3]。
E.coli HCP與CHO HCP有較大的差別
大致可以分為以下幾類:
1、黏附素(Adhesins):Curli (CsgA) ,形成淀粉樣纖維,促進(jìn)生物膜形成;
2、毒素(Toxins):α-溶血素(HlyA), 孔形成毒素,溶解紅細(xì)胞和上皮細(xì)胞;
3、侵襲素(Invasins):OmpA, 促進(jìn)E. coli侵襲血腦屏障(NMEC),抵抗補(bǔ)體殺傷;
4、免疫逃逸相關(guān)蛋白: LPS(脂多糖), 誘導(dǎo)炎癥,但逃避免疫識別;
5、分泌系統(tǒng)效應(yīng)蛋白:EspC(自轉(zhuǎn)運(yùn)蛋白酶),破壞上皮屏障;
6、宿主互作效應(yīng)蛋白:NleH, 宿主NF-κB通路,減少炎癥;
7、噬菌體/質(zhì)粒編碼毒力蛋白:Colibactin, 基因毒素,誘導(dǎo)DNA損傷。
Jones等報(bào)道,殘留的HCP不僅會(huì)有免疫原性,而且還有藥物輔料降解的功能。對宿主菌/細(xì)胞高風(fēng)險(xiǎn)因子基因做基因敲除,或使用空載HCP模擬藥物生產(chǎn)和純化流程,如深層過濾、親和純化、陰陽離子純化產(chǎn)物中的HCP殘留分析尤為重要。在優(yōu)化藥物優(yōu)化純化過程中,不乏有較難去除的HCP。Zhu等報(bào)道過,E.coli表達(dá)的malaria vaccine在純化過程中的HCP含量范圍在90 ng or <1100 ppm/劑。但是臨床能接受不形成慢性疾病的HCP含量范圍僅僅<100 ppm/劑[4,5,6]。
所以,對HCP需要應(yīng)檢盡檢,防止漏檢,不斷提升檢測靈敏度和覆蓋度。
